“Aims:

To develop total DNA extraction protocol from saline soil for further metagenomic applications.

Methods and Results:

The protocols combine the application of mechanical (Beads and Sonicator) and Soft Lysis (SDS and enzymes) method for the isolation of total DNA from saline soil of coastal Gujarat followed by its quantification and purity assessment. The quality and purity of metagenomic DNA was quite consistent and reliable, although it contained

residual concentartions of humic acid. The extracted DNA was used to successfully amplify 16S rRNA mTOR inhibitor region. The amplicons were suitable for further applications such as diversity-based analysis by denaturing gradient gel electrophoresis (DGGE).

Conclusions:

The methods appear to have

wide applicability in investigating molecular diversity and exploring functional genes from the total DNA.

Significance and Impact of the Study:

Subcutaneous administration of morphine (0.5, 1, 2.5, 5, 7.5

and 10 mg/kg) induced CPP only at the doses of >= 5 mg/kg. The dose of 0.5 mg/kg of morphine was selected as the appropriate (ineffective) dose for induction of CPP in Galunisertib animals which were previously received AM251 (5, 25 and 125 ng/0.5 mu l per side) once daily for three days as a sub-chronic administration or those that received a single dose on the test day. Bilateral intra-accumbal sub-chronic but not single administration of AM251 dose-dependently produced sensitization to morphine and induced CPP by ineffective dose of morphine (0.5 mg/kg) in the rat. Bilateral intra-accumbal administration of neither saline nor DMSO (0.5 mu l/side) had effects on sensitization to morphine.
